Biphasic bioprocess cultivation of alkaline phosphatase-expressing CHO cells in chemically defined, protein- and peptide-free ChoMaster® minimal media at pilot scale, and methods for the use of ChoMaster® minimal media for recombinant protein expression in single-use bioreactors.

Katharina Blaschczok and her team at the Institute of Biotechnology of the Zurich University of Applied Science recently published a short communication on the biphasic extended batch cultivation of CHO cells with ChoMaster HP-1 and HP-5 minimal media at pilot scale in Engineering in Life Sciences. (2014), vol. 0, 1-5 (DOI: 10.1002/elsc.201300069, Wiley-VCH Verlag). Recombinant alkaline phoshatase-secreting CHO cells adapted to proliferation in such media were induced by dual medium exchange – growth medium replaced by production medium – and temperature shift in ChoMaster media.

Moreover, a comprehensive list of material and methods for inoculum preparation and recombinant protein expression in single-use bioreactors was published by Eibl et al. in Ralf Poertner’s Animal Cell Biotechnology: Methods and Protocols, Methods in Molecular Biology (2014), vol. 1104, 265-284 (DOI: 10.1007/978-1-62703-733-4_18, Springer Science + Business Media).
